This was one thing where I wasn't sure if I agreed with Fury's Big Week, it had Coulson very involved in the development of the Destroyer gun but in the film, he has access to the gun but does seem surprised at what it does/effectiveness. It makes me think he wasn't that involved and just knew of it since he's the Marvel version of Ianto and knows everything.
